Advanced Tomography
The Advanced Tomography software is an optional plug-in to Gatan’s DigitalMicrograph product. Together with the Gatan TEM Tomography software the Advanced Tomography product enables tomography tracking to be carried out away from the data acquisition area and under different illumination conditions.
During the acquisition of a Tomography tilt series you typically need to track movements of the stage in X, Y, and Z, and this involves acquiring additional images to use in the calculations. Sometimes you do not want to expose the interesting area of the specimen to the increased dose that this would entail, or perhaps the tracking would perform better on another region. The Advanced Tomography software allows you to set up the X/Y and Z tracking areas at locations displaced along the tilt axis from that of the data acquisition area. At these locations you can also choose different illumination conditions (spot size and brightness) than those at the data acquisition site. As a result the area of interest receives minimum dose and the tracking images can be of high signal-to-noise ratio on perhaps a more contrast-rich part of the specimen.
The Advanced Tomography software integrates seamlessly with the Gatan TEM Tomography software. A new tomography mode is added to the Tomography palette and a new tab is added in the Tomography setup dialog. There you can adjust the offsets of the acquisition, X/Y and Z tracking states along the tilt axis, view a sample of the images that will be acquired in each state and adjust the illumination conditions (spot size and brightness) to use at each location. While you make these adjustments you can watch the positions of the three states on a schematic overview of the sample.
During the tilt series acquisition the Tomography software will switch between the acquisition, X/Y and Z tracking states as usual but now they may spatially separated and feature different illumination conditions. You can ensure that the area of interest is exposed to the minimum dose.
